The Cardinals picked up an 11-5 victory on Wednesday at Jim Patterson Stadium.
Louisville (13-4) has now won nine of its last 10 games and is 12-2 during its current homestand.
The Cardinals jumped on the board with a pair of runs in the first and then added another on a run-scoring double from Dalton Rushing in the third.
Bellarmine (2-15) cut the lead to just one with a two-run homer in the top of the fourth, but Louisville answered right back with three runs of its own in the bottom half of the frame.
A throwing error and a wild pitch allowed two runs to come home and Cameron Masterman picked up the first of his three RBIs with a sacrifice fly. Masterman made it a three-RBI day in the eighth, doubling into right-center to score two.
Rushing and Jack Payton added insurance runs with run-scoring hits of their own in the eighth to help the Cardinals reach double digits in the run column for the seventh time this season and third straight game.
Luke Seed (1-0) pitched four innings in his first start of the season, allowing just the two-run homer on his way to his first victory as a Louisville Cardinal. Ben Wiegman retired six of the seven batters he faced in two scoreless innings and Michael Prosecky tossed two scoreless of his own for his fifth save of the year.
Masterman, Rushing and Payton each had two hits to lead the Louisville offense. Masterman drove in three runs for the second consecutive day.
The Cardinals open up ACC play this weekend with a three-game set against No. 1 Notre Dame. The series is scheduled to begin Friday at 7 p.m. ET at Jim Patterson Stadium.
